Biofouling Vol.26,No.3,April2010,301312 Theuronicacidsassay:amethodforthedeterminationofchemicalactivityon biolmEPS KristinaD.A.MojicaandMichaelJ.Cooney DepartmentofOceanography,SchoolofOceanandEarthSciencesand Technology,UniversityofHawaii,Honolulu,HI96822,USA;bHawaiiNatural EnergyInstitute,SchoolofOceanandEarthSciencesandTechnology,Universityof Hawaii,Honolulu,HI96822,USA (Received7June2009;nalversionreceived20November2009) Inthiswork,theuronicacidsassaywasevaluatedforitspotentialtofunctionasa bioassaytoscreenforantagonisticactivitygainsttheproductionofmicrobialbiolm exopolysaccharide(EPS).Theassaywasrstappliedtobiolmsproducedinthe presenceoftwouniversaldisinfectants(sodiumhypochloriteandsodiumdodecyl sulfate)knownoinhibitmicrobialgrowthandbiolmformation.Theperformanceof theassaywasthecharacterizedthroughstatisticalassessmentofthreshold concentrationsfordisinfectioneciencyandconsistencyrelativetovaluesreportedin theliterature.Theassaywasthenevaluatedforitsutilityinscreeningforenzymatic orchemicalinhibitorsofbiolmformation(egglycosidases,halogenatedfuranones, andsemi-crudefractionsextractedfromminimallyfouledmarineplants)andits abilitytodistinguishbetweentrueanti-biolmactivityandsimpledisinfection.
