几种疾病外周血单核细胞对

OK-432增殖反应的比较

[摘要]　目的　研究掌跖脓疱病(pustulosispalmarisetplantaris,PPP)、寻常型银屑病(psoriasisvulgaris,PV)、泛发性脓疱型银屑病(generalizedpustularpsoriasis,GPP)、连续性肢端皮炎(acrodermatitiscontinua,AC)患者的外周血单个核细胞(peripheralbloodmononuclearcells,PBMC)对白介素-2(interleukin-2,IL-2)或链球菌粉干制剂OK-432的增殖反应,探讨几种疾病的关系。方法　分离提取PPP、GPP、PV、AC及健康患者PBMC,分别加入IL-2或OK-432,第6天MTT法检测细胞增殖活力并测定DNA含量。结果　PPP、GPP、PV、AC患者与健康对照组的PBMC对IL-2的增殖反应相比较无统计学差异(P>0.05)。而GPP、PV、AC患者的PBMC对OK-432呈低增殖反应,与健康对照组或PPP组有统计学差异(P<0.01)。PPP患者的PBMC对OK-432呈高增殖反应,与健康对照组无统计学差异(P>0.05)。结论　PPP和健康对照组对OK-432的反应相似,而与其它几种疾病反应不同,提示PPP与其它几种疾病可能存在不同的发病背景。

Abstract:Objective　Tofindthedifferentresponsesofperipheralbloodmononuclearcells(PBMC)tointerleukin-2(IL-2)orOK-432amongpustulosispalmarisetplantaris(PPP),psoriasisvulgaris(PV),generalizedpustularpsoriasis(GPP)andacrodermatitiscontinua(AC)invitroandtoexploretherela-tionshipsamongthediseasesabove.Methods　PBMCwereobtainedfromthesepatientsandhealthycontrols,thenIL-2orOK-432wasaddedinthemandtheproliferationactivityandDNAcontentwereas-sayedatthesixthday.Results　WhenthePBMCwerestimulatedwithIL-2,nosignificantdifferencewasfoundinproliferationresponsesbetweenpatientswithPPP,PV,GPP,ACandhealthycontrols.However,PBMCfromPV,GPP,andACpatientsshowedsignificantsmallerresponsestoOK-432thanthosefromthehealthycontrolsandPPP.Conclusion　ThedifferentresponsestoOK-432betweenPPPandPV,GPP,ACsuggestthatPPPhasadifferentpathogeneticbackgroundfromthatunderlyingPV,GPP,andAC.
